Abstract

OBJECTIVE: To optimize the extraction process of crude polysaccharides from leaves and improve the extraction rate and efficiency. METHODS: The ketone-sulfuric acid method was used to detect the effects of decoction, ultrasonic and microwave methods on the extraction rate of crude polysaccharides from leaves. The single factor and positive were used in water and alkaline water (NaOH solution). The experiment was carried out. The extraction rate of crude polysaccharide was used as the index. The effects of water addition, NaOH solution concentration, extraction temperature and extraction time on the extraction of crude polysaccharide by alkaline water were studied. The process parameters were optimized and verified. Results: The alkaline water ultrasonic method was superior to other extraction methods; the optimal process parameters were 0.3 mol/L NaOH aqueous solution with 30 times of medicinal material and 70 min ultrasonic treatment at 50 °C. The verification results showed that the average extraction rate of crude polysaccharide was 25.76%. (RSD = 3.51%, n = 6). Conclusion: The preferred alkaline water ultrasonic method can extract the crude polysaccharide components from the leaves.

Conclusion

The influence degree of each factor on the extraction rate of crude polysaccharide was as follows: water addition amount (A), solvent concentration (B), extraction temperature (C), extraction time (D), and the optimal process parameter was determined as A2B2C2D2, that is, the water addition amount was 30 times. The concentration of NaOH aqueous solution was 0.3 mol/L, the extraction temperature was 50 ° C, and the extraction time was 70 min.

In actual production, the extraction method of the boiling method generally uses an extraction tank and is heated by steam, so the energy consumption is large; at the same time, the production process is affected by the steam output of the boiler, thereby restricting the production process. The ultrasonic extraction energy consumption is low, the process independence is good, and other factors have little interference. When the extraction rate is close, the production efficiency advantage is obvious. Today's Chinese medicine extraction technology develops rapidly. With the application of large-scale ultrasonic equipment in extraction, this method will gradually exert its advantages in large-scale industrial production, so it has a good promotion value.
