Abstract

The physin sterol is present in a large amount in the physalis seed oil, and the seed oil is extracted therefrom by microwave assisted method, and then saponified and extracted to obtain an acid sterol. The response surface method was used to optimize the extraction conditions of phytochemical sterol. The results showed that: under the condition of absolute ethanol as extractant, the liquid-to-liquid ratio was 11:1 mL/g, the extraction time was 7 min, the extraction temperature was 51 °C, the microwave power was 500 W, and the physalis was used. The yield of sterol was 1.771 mg / g. The original acid pulp seeds and ethanol microwave-assisted extraction of physalis seeds were scanned and observed by transmission electron microscopy. It can be seen that the phytoplasma seed cells have a large amount of fat particles on the surface and the particles are full; the microwave-assisted extraction of the physalis seed cell wall is completely broken. The cell membrane permeability is increased, and a large amount of seed oil is leached from the cells. The microwave-assisted method has a good effect on the extraction of phytochemicals.

Microwave-assisted extraction method was used to study the process conditions of microwave-assisted extraction of citric acid 甾287 alcohol by single factor and response surface analysis. The results showed that: anhydrous ethanol was used as extractant, liquid. The ratio of material ratio was 11:1mL /g, extraction time was 7 min, extraction temperature was 51 °C, microwave power was 500 W. After evaporation of ethanol, KOH was added to a concentration of 1.5 mol / L in KOH ethanol solution for 1 h. Petroleum ether was extracted and petroleum ether layer was taken. The solvent was evaporated to dryness to obtain an acid phytosterol extract. The yield of sterol was 1.771 mg / g, which was basically consistent with the theoretical prediction. The microwave-assisted extraction mechanism was carried out by microwave-assisted extraction of physalis seeds from ethanol and ethanol. The mechanism of microwave-assisted extraction was caused by microwave radiation and thermal effects. The cell wall was completely broken and cell permeability was observed. Increasing the dissolution of internal dissolved matter accelerates the dissolution rate of the sterol sterol.

Instrument

Weigh 6 g of physalis seed powder into a three-necked flask, add a certain amount of extraction solvent, and extract it on a microwave extractor. After the extraction, the solvent is evaporated to obtain yellow physalis seed oil; the concentration is 1.5 mol / The KOH ethanol solution of L is saponified for 1 h, extracted with petroleum ether, and the petroleum ether extract is rotatized to complete evaporation, and the solvent is recovered to obtain a crude extract of physalis sterol. The volume is adjusted with glacial acetic acid, and a certain amount of sample liquid is taken for determination. The acid sterol content was calculated and each sample was averaged three times in parallel.
